Strains of stated genotype were crossed and meiotic progression followed using DIC/Nomarski to show location of ascus and NLS-GFP to show nucleus. NLS-GFP is shown in green and DIC/Normarski is in red. Images collected are flat projections of serial Z-sections collected every 5 min. Playback rate is 10 frames/sec. (For details see Materials and Methods.)

• video 1 wild type (1.06 MB) Cross of two wild-type haploids. Two distinct meiotic divisions occur to produce four equal sized GFP signals in the lower ascus. Movie ends prior to completion of meiosis in upper ascus. Total elapsed time was 6 hours 10 minutes.
• video 2 rdh54 (3.67 MB) Cross of two rdh54∆ haploids. In this example, rdh54∆ does not greatly disrupt divisions and four nearly equal sized GFP signals are produced. However, the first meiotic division is unequal. Total elapsed time was 4 hours 35 minutes.
• video 3 rdh54 (2.56 MB) Cross of two rdh54∆ haploids. In this example, rdh54∆ results in abberant divisions and greater than four GFP signals appear after the divisions. Note the nuclear GFP signal left at midplane after first division. Total elapsed time was 5 hours 5 minutes.
• video 4 rdh54 (2.77 MB) Cross of two rdh54∆ rhp54∆ haploids. The first meiotic division is quite abberant, with nearly all of the nucleus segrating upward; only a very small GFP signal moves to the opposite end of the ascus. A second meiotic division ensues resulting in five GFP signals. Total elapsed time was 5 hours 5 minutes.
• video 5 rhp54 (845 kb) Cross of two rdh54∆ rhp54∆ haploids. Movie begins at MI in which a large portion of nuclear GFP signal remains a midplane. the second meioitic division results in highly aberrant meiotic products. Total elapsed time was 2 hours 20 minutes.
