  • Fig. 1S. Nuclear import of c-Fos is only required for 4 h to trigger neuritogenesis. Cytoplasmic c-Fos expression is required at all times of neurite growth. A time course of the effect of NLSP or NLSP + ASO addition on neurite outgrowth has been performed between 0 and 16 h of NGF treatment. For this, cells cultured with NGF for 4 days were subjected to different temporal additions of NLSP or NLSP + ASO as indicated at left. After 4 days in culture, cells were immunostained for c-Fos (left panels) and α-Tub (right panels). Morphometric quantification data obtained as in Fig.1 are posted to the right of the corresponding row. As can be observed in the figure, nuclear c-Fos is required to trigger neuritogenesis only up to 4 h of NGF treatment whereas blocking cytoplasmic c-Fos expression results in cells in which neurite not only stop growing, they also retract. Bar, 10 µm.
