Rab14 Is Involved in Membrane Trafficking between the Golgi Complex and Endosomes

GFP-Rab14-wt and –Q70L positive structures are dynamic, whereas GFP-Rab14-S25N is localized to Golgi stacks and static. NRK cells were transfected with GFP-Rab14-wt, GFP-Rab14-Q70L or GFP-Rab14-S25N constructs. After 20-24 hrs, the GFP movies were collected at 25 °C using Olympus IX-70 Deconvolution microscope (Plan Apochromat 60x/1.4 NA, oil immersion objective lens) for wild type (70 frames, 2 sec interval per frame), Q70L (42 frames) and S25N (40 frames). As shown in movie 1A (Rab14-wt) and movie 1B (Rab14-Q70L), the Rab14 positive vesicles and tubules are dynamic in nature. In contrast, the Rab14-S25N label is mostly confined to Golgi stacks and static (movie 1C). GFP-Rab14-wt transfected cells were incubated with Alexa 594 labeled transferrin, after 10 min uptake, the GFP movie (35 frames) was recorded (movie 1D).
