Histograms show the percentage of siRNA-treated cells expressing CHO1 proteins at three different levels. <_1: equal to or less than endogenous CHO1 (100% = 1) in control cells, 1-3: between 100-300%, and >_3: over 300% of endogenous CHO1 in control cells. Fluorescence intensity of polyclonal anti-CHO1 antibodies was measured in 30 prometa/metaphase cells expressing different CHO1 constructs using the ImagePro Plus image analysis software package and compared with that of 30 control and mock-rescued cells at prometa/metaphase stages. Expression levels are comparable among all twelve rescue constructs.

Histograms show the percentage of siRNA-treated cells expressing different rescue constructs probed by anti-HA antibodies. The protein amount was quantified using the ImagePro Plus program as arbitrary units of HA fluorescence which were divided into three categories of low (9-40 units), medium (41-70 units) and high (71-100) expression. The level of protein expression is comparable among twelve different rescue constructs.
