Movie1Control.mov (5.12 MB)
-
Time-lapse phase contrast sequence of a Swiss 3T3 fibroblast subjected to shear flow (τw= 9.4 dyn/cm2). Images were captured every minute to document nucleus motion (rotation and translocation). The arrow indicates the direction of flow. Bar, 20 μm.

Time-lapse phase contrast sequence of a Swiss 3T3 fibroblast transfected with EGFP:Cdc42T17N subjected to shear flow (τw= 9.4 dyn/cm2). Images were captured every minute to document nucleus motion. The arrow indicates the direction of flow. Bar, 20 μm.
Movie5NocodazoleTreated.mov (5.67 MB)
-
Time-lapse phase contrast sequence of a Swiss 3T3 fibroblast treated with nocodazole subjected to shear flow (τw= 9.4 dyn/cm2). Images were captured every minute to document nucleus motion. The arrow indicates the direction of flow. Bar, 20 μm.
Movie6Nucleliof5Conditions.mov (9.05 MB)

Montage of nuclei of cells shown in Movie 1-5. In control, EGFP:RhoAT19N, and EGFP:Rac1T17N transfected cells, rigid-body translocation and minor rotation occur. EGFP:Cdc42T17N transfected cell exhibits enhanced rigid-body movement and dramatic concerted NR. In nocodazole-treated fibroblast, the nucleus shape is significantly compromised and nucleoli movements are independent from each other. Bars, 20 μm.
