4D imaging showing events associated with retention of mitochondria in the bud. Wild-type yeast expressing CS1-GFP (BY4741) grown to mid-log phase were monitored by 4D imaging (time-lapse microscopy combined with 3D reconstruction). Z-sections through the entire cell were obtained as described above. The interval between each Z-series was 5 sec. The first segment of the movie shows a rotation of a 3D projection of GFP-labeled mitochondria in the cell at the onset of imaging. The second segment shows a time-lapse series of 3D reconstructions that were projected into a single image. The movie illustrates linear movement of mitochondria towards the bud tip, and immobilization of mitochondria in the bud tip.
