(A) Conservative substitution of leucine 252 to isoleucine did not affect the basolateral targeting of CD147 as seen in confocal xy and xz sections. (B) The GFP-tag at C-terminus of CD147 does not affect its trafficking. Steady-state domain-specific biotinylation shows basolateral targeting of CD147(WT)-GFP and apical localization of CD147(L252A)-GFP. Clones expressing similar levels of both proteins were used for the targeting assay.
