Movies were generated from images obtained on a Zeiss LSM 510 confocal microscope with 100X objective.

This article contains the following supporting material:

• movie1.mov - Ax2 cells expressing the VwkA-GFP construct were imaged after addition of FM2-10 dye, with frame collection initiating within ~20 sec of dye addition, and frames collected at 10 sec intervals. As in figures in manuscript, FM2-10 signal is collected in the red channel, GFP image in the green channel, and yellow represents areas of overlapping signal.
  
  
• movie2.mov - All parameters as in movie VwkA-GFP-FM-1.mov.
  
  
• movie3.mov - Ax2 cells expressing the VwkA-GFP construct were fixed as described in Methods and a Z-series of optical slices was collected to create the movie. Punctate localization of VwkA-GFP throughout cytoplasm is apparent, as is the much brighter punctate perinuclear signal that is present in most cells.
