HeLa cells that stably express Grb2-YFP and lack endogenous Grb2 (Cl-1, see Figs. 1-2) were maintained in the microscope environmental chamber at 33°r;C. The time-lapse imaging was performed through YFP filter channel. EGF was added at time ?0? (first frame). The elapsing time is shown in minutes above the image. The appearance of Grb2- YFP dots at the periphery of the cell which probably corresponds to the recruitment of Grb2-YFP to coated pits was observed starting with the second frame of the movie. At later times, a large number of endosomes containing Grb2-YFP is seen in the perinuclear area. The exposure integration times were 200 ms.
