Supplementary Material
Figure S1. Pmh1 is an essential protein homologous to Mat1. (A) Alignment of Pmh1
with Tfb3 and Mat1. (B) Tetrad dissection of pmh1+/pmh1::ura4+ strain produced two
viable and two inviable spores. (Tetrad 3 had only one viable spore.) (C) pmh1? spores
were germinated in selective media (-uracil), grown for 48 h, fixed and DAPI-stained.
Cells divided ~six times and arrested with two nuclei and a single division septum, with a
minority of cells containing four nuclei and three division septa.
Figure S2. Global gene expression changes in mcs6 and pmh1 mutants. Histograms
showing changes in signal ratios (compared to signal at time 0) in mcs6ts1 and pmh1-26
mutants after incubation for various times at restrictive temperature, as indicated,
normalized with reference to external RNA spikes. The average and median signal ratios,
and the standard deviation for each condition are as follows: pmh1-26 at 0.5 h (avg. 1.08,
med. 1.06, s.d. 0.22); pmh1-26 at 2 h (0.77, 0.75, 0.21); mcs6ts1 at 0.5 h (0.82, 0.77, 0.35);
mcs6ts1 at 2 h (0.74, 0.66, 0.46).
