Southern blot analysis of relative mtDNA copy number of the same strains shown in Figure 1. The relative mtDNA copy number (plotted on the ordinate) of a pif1 null (pif1??), rrm3 null (rrm3??) and the corresponding double-mutant (pif1?? rrm3??) strain is shown compared to that of the isogenic wild-type (wt) strain DBY2006. The Southern blots were performed using probes for mtDNA (COX3) and nuclear DNA (ACT1) as described (Rodeheffer, M. S. and Shadel, G. S. 2003. J. Biol. Chem. 278, 18695-18701).
