Dextran loading of GFP-PipB2 transfected HeLa cells. HeLa cells were transfected for 24 hr with GFP-PipB2 and then incubated with Alexa Fluor® 568-conjugated dextran for 90 min followed by a 30 min chase. Images were acquired at 600 ms intervals. GFP-PipB2 is green and dextran is red. Speed is 5x actual. A GFP-PipB2 transfected cell is at the bottom of the image and an untransfected cell is at the top for comparison. Peripheral accumulation of GFP-PipB2 and dextran is evident. Note the dynamic nature of the GFP-PipB2 vesicular-tubular structures and their bidirectional movement. Inset in top left hand corner is 2x enlargement of GFP-PipB2 transfected cell. Small dextran-positive endosomes are seen to move along GFP-PipB2-positive vesicular-tubular structures. Large accumulations of dextran are surrounded by GFP-PipB2-positive membrane.
