Video 1

Video1 corresponds to Figure 1. Volume-rendering View of Centromeres in the Nuclei of Guard Cells. Centromeres were labeled with HTR12-Venus in green, and chromatin was labeled with H2B(HTB1)-CFP in red. All twenty centromeres in a pair of guard cells were observed to localize at the nuclear periphery.
Video 2
-
Video2 corresponds to Figure 4A. Dynamic Views of Centromeres in a Root Meristematic Cell in Interphase. The centromeres are labeled with HTR12-GFP. Time-lapse 2-D images (single section) up to 75 minutes were assembled into the video.
Video 3
-
Video3 corresponds to Figure 5A. Dynamic views of Centromere Movements through Mitosis. The centromeres were labeled with HTR12-GFP in green.
Video 4
-
Video4 corresponds to Supplementary Figure S3A. Dynamic views of Centromere Movements through Mitosis. The centromeres were labeled with HTR12-GFP in green.
Figure S1
-
Twenty image sections with 0.2 μm interval through nuclei of guard cells. The centromeres are labeled with HTR12-Venus in green, and chromatin lablled with HTB1-CFP in magenta. Centromeres in the nuclei, which were defined as brightest point at the local foci along the z-axis, are highlighted with arrows. Bar = 5 μm.
Figure S2
-
Seven image sections with 0.6 μm interval through nuclei of mother cell and its daughter cells. The centromeres are labeled with HTR12-GFP. Due to the lower resolution in the z-axis, some centromeres are stretched along the z-axis. Centromeres in the nuclei, which were defined as brightest point at the local foci along the z-axis, are highlighted with numbered arrows. Bar = 5 μm.
Figure S3A
-
Tracking Centromeres through Mitosis in 3-D (see also Supplementary Video4).
(A) Projection images of nuclei of a mitotic cell at elapse time of 0 min, 6 min, 19 min 30 s, 36 min 45 s and 48 min. Centromeres are highlighted with numbered arrows at each time point. Cycled spots are projected signals from adjacent cells. Bar = 5 μm.
Figure S3B

(B) Diagram of positions of centromeres in nuclei of mother cell and two daughter cells. Z-axis is directed to the bottom of the nuclei. Centromeres in the top sections of the nuclei are represented by numbered dark gray-scaled dots, and centromeres in the bottom sections of the nuclei are represented by numbered light gray-scaled dots.
