Supplementary Figure -
A. Activation of the mitotic checkpoint after Dgrip84 depletion in S2 cells.Cells were stained for BubR1 (green signal). In control cells, the chromosomes that are aligned along the metaphase plate show a weak signal, while isolated chromosome that has not yet reached the metaphase plate exhibits intense signal at the two kinetochores (arrowhead). In all cases, RNAi-treated figures present a strong BubR1 accumulation at the kinetochores. B. Characterization of the poles after Dgrip84 depletion. The staining in green corresponds to the polar markers Dgrip84 (a,b,c), γ-tubulin (d,e,f) and Dgrip91 (g,h,i). Note that in treated cells, neither Dgrip84 nor γ-tubulin staining could be detected at the poles and along spindle microtubules. Scale bars, 5 μm ; Aa-c and Ba-h except d, same scale bar than in Bi.
