• Figure 1 - Demonstration of TGF-β induced Smad-independent signaling. (A) Activation of the MEK pathway was confirmed via immunoblotting for p-ERK1/2, with an increase in protein levels upon stimulation with TGF-β1 for 24 hours. This increase was prevented upon treatment of cells with a MEK1/2 inhibitor (U0126). (B) The PI3K pathway was also confirmed by immunoblotting with p-AKT antibody, showing a sharp increase after 24 hours of stimulation with TGF-β1. Increases in p-AKT were prevented in cells treated with a PI3K inhibitor (LY294002).

• Figure 2 - Confirmation of TGF-β1 induced transition of epithelium to mesenchyme. Increases in Vimentin expression were observed by immunocytochemistry 72 hours post treatment with TGF-β1.
