• Supplemental Figure 1 - Actin clump accumulation of Scd5p in ark1Δprk1Δ cells depends on the 9R region.

  ark1Δ prk1Δ cells (SL3799) expressing GFP-Scd5p, GFP-Scd5p-Δ3R, GFP-Scd5p-Δ9R, GFP-Scd5p-ΔCT, and GFP-Scd5p-ΔCT+NES were grown to log-phase at 30°C, fixed with formaldehyde, and stained with Alexa-594-phalloidin. GFP or actin signals were visualized in the FITC (green) or CY3 (red) channels, respectively. Merged images with yellow indicating co-localization are also shown.

  Cultures of cells expressing baits, pGBDU (empty vector; GBD), pNT1 (WT; pGBD-scd5-Δ338/ΔCT), pJSC54 (NLS; pGBD-scd5-ΔCT-NLS), or pJSC19 (PP1Δ2; pGBD-scd5-ΔCT-PP1Δ2), and preys, pGAD (empty vector) or pGAD-GLC7, were spotted on selective medium lacking leucine and uracil (C-LEU-URA) to monitor growth or medium lacking leucine, uracil, and adenine (C-LEU-URA-ADE) to assay for two hybrid activation of the GAL2:ADE2 reporter. Plates were grown for 3 days at 30°C.
