Dynamics of HDAC6-GFP during PBL migration over a FN substratum. Time lapse video sequence of HDAC6-GFP-nucleofected-PBL migration, on a 50 μg/mL FN substratum, during a total observation time of 8 min. The maximum projection of the informative sections from the GFP confocal stack is merged with differential interference contrast (DIC) images. Green color corresponds to the GFP emission and the halftones in blue are the DIC image. HDAC6-GFP was localized at the leading lamellipodium, and in lesser extension at the rear pole.

Dynamics of GFP alone during CEM cell migration over a FN substratum. Time lapse video sequence of pLZR IRES GFP-transduced-CEM leukemic T cell migration, on a 20 μg/mL FN substratum, during a total observation time of 8 min. Six confocal sections were taken every 20 s. The maximum projection of these confocal sections from the GFP signal is presented. The fluorescence intensity of GFP, that represents the concentration of expressed GFP, is depicted in an arbitrary gradient pseudocolor map (from intense blue for the minor to deep brown for the maximal intensity signal). GFP is evenly distributed in cell body.

Dynamics of HDAC6-GFP during CEM cell migration on FN. Time lapse video sequence of pLZR IRES HDAC6-GFP transduced-CEM leukemic T cell migration on FN (20 μg/mL), during a total observation time of 21 min. The maximum projection of the six sections captured every 20 s from the GFP confocal stack was taken to compose the image. The fluorescence intensity of GFP, that represent the concentration of expressed HDAC6-GFP, is depicted in the arbitrary gradient pseudocolor map. HDAC6-GFP was clearly concentrated at the leading lamellipodium (brown color), and in lesser extent at the rear pole (yellow-green).
