Figure 1 -
Stimulation of SPR-Hela cells with substance P or TPA does not change the basal membrane localization of Epsin, Eps15 or Dab2. SPR-Hela cells were stimulated with either 100 nM substance P or 0.5 μM TPA for 3 minutes. Following fixation in 4 % PFA and permeablizing with saponin, the cells were stained with (A) goat anti-Epsin (Santa Cruz Biotechnology Inc.), rabbit anti-Eps15 (C-20, Santa Cruz Biotechnology Inc.) and (B) mouse anti-Dab2 (BD Transduction Laboratories). For each experiment, stimulated and unstimulated cells were also stained with anti-NbC to control for the efficacy of stimulation (bottom panels in A and B). Primary antibodies were visualized using either Al488- or Cy3-labelled secondary antibodies. Size bars indicate 10μm.
