Hog1p may act through Rck2p to mediate arsenic tolerance.
(A) The rck2Δ mutant is As(III) sensitive. Growth of wild type, hog1Δ, rck2Δ and hog1Δrck2Δ was monitored in the presence of increasing concentrations of As(III). Cells were incubated in minimal medium at an OD600 of 0.05-0.10 and grown in the presence of the indicated As(III) concentrations. The OD600 of each culture was determined after 24 h and plotted on the graph. The experiments were done in triplicate and error bars represent the S.D.
(B) RCK2 overexpression partially suppresses the As(III) sensitivity of hog1Δ. Wild type and hog1Δ cells were transformed either with an episomal plasmid containing RCK2 (pRCK2) or an empty control vector (pRS426) and growth of the transformants was monitored in the presence of As(III) as above. The error bars represent the S.D.
