Supplemental Figure 1 -
Ectopic expression from the MET3 promoter of either BEM3 or RGA2 rescues the characteristic phenotypes of the bem3Δ/Δ rga2Δ/Δ mutant. When growing under pseudohyphal conditions bem3Δ/Δ rga2Δ/Δ buds are elongated and lack constrictions at the mother cell neck. Expression of either BEM3 or RGA2 from theMET3 promoter restores constrictions to the bud neck and results in shorter and broader buds. BEM3 or RGA2 was reintroduced into the bem3Δ/Δ rga2Δ/Δ strain under the control of the MET3 promoter strain using the pCaEXP vector which integrates at the RP10 locus and uses URA3 as a selectable marker. (A-C) Stationary phase cells of the indicated genotype were reinoculated into SD medium of pH 6.0 and incubated at 36ºC for 100 minutes before samples were removed and fixed in 2.5% formaldehyde for 10 minutes.
