Inhibition of the PI3-K/Akt pathway does affect cFos induction in PEA-15 overexpressing cells Inhibition of the PI3-K/Akt pathway does affect cFos induction in PEA-15 overexpressing cells. Stable clones of CHO-AT1A cells expressing GFP and GFP-PEA-15 were treated or not with 10 μM LY294002 for 30 min and stimulated with 100 nM AngII for 1 hour. Cell lysates were prepared with 1% SDS. 50 μg proteins were separated by SDS-PAGE and immunoblotted with anti-cFos antibody.

Inhibition of the PI3-K/Akt pathway does affect ERK1/2 localization in PEA-15 overexpressing cells Inhibition of the PI3-K/Akt pathway does affect ERK1/2 localization in PEA-15 overexpressing cells. Serum-starved clones of CHO-AT1A cells expressing GFP, GFP-PEA-15 or GFP-D74A were pretreated or not with 10 μM LY294002 for 30 min. Subsequently, cells were stimulated with 100 nM AngII for 3 hours (AngII, AngII + LY, respectively) or maintained in serum-free medium (Control, respectively). Cells were fixed with cold methanol and ERK immunoreactivity was detected using a polyclonal anti-ERK1 antibody (red). GFP fluorescence was directly imaged (green)
