Supplemental Figure 1
-
Emission spectra of HE oxidation in intact MPMVECs. O2.- generation following inhibition of mitochondrial complex III by antimycin A (AA) or following extracellular O2..- addition to intact HE-loaded MPMVECs resulted in similar emission spectra when excited at 494 nm. The fluorescence signal was significantly elevated in treated compared to untreated cells. A baseline tracing (buffer) in the absence of cells is shown.
Supplemental Figure 2
-
Receptor-mediated endothelial O2..- generation. Representative images of HE-loaded HPMVECs stimulated with Angiotensin II (Ang II; 2 μM) or Thrombin (0.5 U/ml) for 1 hour under control conditions or following pretreatment with DIDS (300 μM) or apocynin (Apo; 2 μM).

Effect of Tg on ΔΨm. Addition of Tg (2 μM) did not trigger ΔΨm alterations as detected by nuclear rhodamine 123 fluorescence. FCCP (2 μM) was added to as a positive control following Tg addition.
