Wild type (A) and icmA- (B) cells expressing N-term PI3K-GFP were pulsed with cAMP and plated on glass-bottomed petri dishes. A burst of cAMP was applied from a nearby pipette connected to a pulse controller. Relocalization of cAMP occurs in both cell types with 6 sec of treatment with cAMP.

Cells expressing GFP-PhdA were pulsed with cAMP and the subjected to bursts of cAMP from a nearby micropipette. Protein relocalization in wild type cells is seen within 6 sec of application of cAMP (A). Most icmA- cells fail to respond (B).
