The density of ANF-GFP containing vesicles at the plasma membrane was the same in cells that expressed wild-type and mutant synaptotagmins (a) Representative footprints of cells that expressed wild-type and mutant synaptotagmins. TIRF images were acquired of live cells resting in basal medium. (b) Dense-core vesicles in footprints (n=20 for each condition) were counted and divided by the total footprint area. The average number of dense-core vesicles per square micron (± SD) was graphed for each cell types.

Ranking of docked complexes. Binding energies for docked complexes with C2A residues within 5Å of SNAP25 D179, D186 and D193 were calculated using the program DoME taking electrostatic and van der Waals potentials into account. Residues of C2A closest to SNAP25 D179, D186 and D193 are indicated for each complex. Only complexes with C2A K196, R199 or K200 were included in the ranking.
