Widefield, deconvolved, and isosurface rendered images of HeLa cells in metaphase (top, middle, and bottom rows, respectively) with microtubules shown in red and DNA in blue. Endogenous HURP (green, left column) colocalizes with chromatin-proximal spindle microtubules. Upon overexpression of GFP-HURP or the microtubule-binding domain of HURP (green; middle and right columns, respectively), the metaphase spindle progressively lengthens and narrows into sharper, hyper-stabilized cones. This microtubule-stabilizing effect of HURP, which is required for the timely assembly of the mitotic spindle and efficient capture of kinetochores, is controlled by phosphorylation by the Aurora A kinase.
